Gonadorelin is a version of gonadotropin-releasing hormone (GnRH). A decapeptide that is created using solid-phase peptide synthesis, It causes the pituitary gland to synthesize and secrete follicle-stimulatinghormone (FSH) and luteinizing hormone (LH), contributing to child development / fertility in adults. The FDA approved gonadorelin for use in women with hypothalamic amenorrhea (absence of menstruation due to a lack of GnRH). Gonadorelin is often used to simulate the physiologic release of GnRHfrom the hypothalamus for patients with delayed puberty or fertility issues caused by hypogonadotropic hypogonadism (a lack of sex hormones due to a lack of a GnRH, FSH and/or LH). It is also used to induce ovulation for women with hypothalamic amenorrhea (absence of menstruation due to lack of GnRH). Gonadorelin administration leads to increased levels of LH and FSH, which then signal the testes orovaries to create and secrete sex hormones.

🔹In Men (for hypogonadotropic hypogonadism, infertility)

SC injections 100–250 mcg Goal: Stimulate LH & FSH secretion to induce testosterone production and spermatogenesis.

 

🔹 In Women (for ovulation induction) Common dose: 100 mcg SC given once to trigger LH surge and ovulation. When: Around cycle day 12–14 (or when the dominant follicle reaches 18– 20 mm, confirmed by ultrasound) • Timing of Intercourse/IUI: Within 24–36 hours after injection. How Often? Single injection during fertile window Typical Uses for Pregnancy: 1. Inducing Ovulation • Used as a trigger to stimulate a natural LH surge, similar to how hCG is used. • One-time injection mid-cycle (around day 12–14), when follicles are mature. • Followed by timed intercourse or intrauterine insemination (IUI).

Important Notes: Gonadorelin may cause multiple births (such as twins,triplets)in patients capable of becoming pregnant. Itis important to discuss the risks and benefits of having  suchpregnancies with your healthcare provider. Gonadorelin maybe used

during the first   trimester ofpregnancy as directedby your healthcare provider. -Monitoring through ultrasounds and blood tests is necessary to assess follicle growth and  hormone levels during the treatment. -In prepubescen tgirls and some people with gonadal function disorders, gonadorelin may cause more FSH to be produced than LH.
